In each episode, with Jameson as the host, five contestants competed in a series of sexual performances for a panel of judges. The judges first eliminated a contestant and then viewers voted for their favorite performer on Playboy. Winners were awarded an exclusive contract with Jameson's movie studio ClubJenna. The show also included interviews with porn actors about each contestant. Ron Jeremy did the lead-in and introduction for the first episodes.

Beginning Friday, November 4, five aspiring adult starlets per episode strut their stuff for Jameson and a panel of celebrity judges in front of a live studio audience to compete for the title of American Sex Star. The winner will be crowned a ClubJenna Girl and will join Ms. Jameson's elite troupe of contract actresses.

In each episode, four contestants compete in a series of sexual performances for the judges and viewers then vote for their favorite performer on Playboy. Winners win an exclusive contract with Jameson's movie studio ClubJenna. The judges will eliminate two contestants and then viewers will vote for who should move on to the finals. Playboy TV's Andrea Lowell appears on season 2 as the Envelope presenter when girls are to be eliminated, or winners announced.
